The Columbian Exchange
A short quote from BBC Radio 4's In Our Time, found via Interconnected.
In 1492, there were 60,000,000 people living in the Americas, which was roughly the same size as the population in Europe and in China. Within a hundred years of first contact with Europeans, fifty five million out of those sixty million were dead.

My own summary after looking into it: infectious diseases such as smallpox and measles were the main cause, with war and enslavement as additional factors.
